A sealed, atmosphere-controlled chamber for live-cell imaging on inverted microscopes. I designed this so I could control the atmosphere for my micro-oxic bacteria. The chamber fits a standard 76 × 25 mm slide and seals shut with a rubber O-ring. The hose barbs allow you to connect a gas source to the chamber so the atmosphere can be defined. This was designed for the Leica DMi8 stage but should be compatible with most inverted stages.
Materials:
2 × rectangular glass coverslips, ~60 × 24 mm, #1.5 thickness
1 × nitrile O-ring, 65 mm ID × 70 mm OD × 2.5 mm cross-section, 70A durometer
6 × M3 × 25 mm hex socket head bolts
6 × M3 hex nuts
Silicone or Tygon tubing for gas lines, sized to fit the barb (~2.4 mm OD shaft, ~2.8 mm flare)
Silicone adhesive for mounting coverslips to printed chamber
